Previous FBI head James the ex-FBI chief has denied the charges to making false statements to lawmakers and impeding a congressional proceeding.
Mr Comey's legal representative submitted the response representing him in a national courtroom in Alexandria, Virginia on mid-week.
Patrick Fitzgerald declared he would attempt to have the case terminated for various reasons including that his defendant, a detractor of former President Trump, was being selectively prosecuted.
Mr Comey was formally accused soon after the former president pressured his attorney general to move forward against him.
During the hearing on mid-week, the defense counsel Patrick Fitzgerald told the magistrate they planned to submit multiple motions to dismiss the case before a court case, claiming the legal action was retaliatory and that a federal prosecutor was unlawfully designated to take over the proceedings.
James Comey's case was originally managed by a Virginia prosecutor, who left his position under pressure from Donald Trump after his probe into an additional political adversary - New York Attorney General Letitia James - failed to initiate prosecution.
Mr Comey appeared in positive mood as he entered the courtroom on Wednesday, conversing with his lawyers and being lighthearted. He was supported by his spouse, Patrice Failor and offspring Maureen Comey, a government attorney who the Trump administration terminated.
After listening to the court recite his rights and the pair of allegations against him in legal proceedings on Wednesday, Mr Comey was inquired if he understood the allegations.
"Absolutely your honour. Thank you very much," he stated to the court.
James Comey was the chief of the Federal Bureau of Investigation from 2013 to 2017 and was fired about several months into Trump's first term as commander-in-chief. At the time, he was overseeing an probe into Russian election interference and whether there were any relationships between the Russian government and the election effort.
While in office, Mr Comey triggered a counter-response from the Democratic party when he announced just shortly prior to the national vote that he was probing political contender Hillary Clinton's use of a non-governmental email account. Charges against Clinton were never brought, resulting in negative feedback from conservative politicians as well.
Government attorneys assert the former director provided false information to Congress during a congressional hearing in recent years when he was being examined about both the previous examination as well as the Russia election probe. They say he deceived the upper house by saying he had not authorized an individual at the Federal Bureau of Investigation to release to journalistic entities data about the agency probes.
Legal representatives also allege Mr Comey of "corruptly endeavoring to influence, hinder and slow" the legislative group by giving misleading testimony to it.
Through a recording the former director published to his Instagram after he was indicted, he declared he was not guilty and accused Donald Trump of conducting himself like a "tyrant".
"Our household have been aware for considerable time that there are consequences to challenging Mr Trump," he commented. "We choose not to live on our bended knee."
"I'm innocent," he stated further. "Thus initiate a court case."
The accusations against the former director surfaced after Donald Trump published on digital channels insisting on his chief prosecutor, Pam Bondi, charge Mr Comey and additional individuals.
